"Captain Francis Xavier 'Frank' Furillo" is in command of the Hill Street Police Station and is the main focus of the show for most of the episodes. He is described as a normal police captain: honest, capable, "by the book," and occasionally irritable. Daniel J. Travanti's performance in Hill Street Blues earned him two Emmys and a Golden Globe. He went on to appear in series such as Poltergeist: The Legacy and Prison Break. His most recent two longest-running performances were in Missing Persons (1993-1994) and Boss (2011-2012), both as "Gerald 'Bab' McGantry." Travanti, like his character on Hill Street Blues, is a recovering alcoholic, as he revealed in an interview with NBC.

"Officer Robert Eugene 'Bobby' Hill" is "Officer Renko's" partner and a decent and loyal guy in general, especially to his partner. Among other things, "Bobby" enjoys boxing and has twice won the "Metro Police Middleweight Championship." After the program ended in 1987, Michael Warren's career did not take off. He has appeared in various programs, including Living Single, Lincoln Heights, Buffy The Vampire Slayer, and, most recently, Anderson's Cross. Warren is the father of four children, two daughters, and two sons. Cash Warren, his son, is married to the well-known actress Jessica Alba.

Bruce Weitz as Det. Michael "Mick" Belker

He frequently discusses societal decay, always guessing about the causes. This occasionally leads to his uttering potentially dangerous things, which he subsequently realizes. James Barrie Sikking went on to appear in a variety of series, including Brooklyn South, Invasion America, and Batman Beyond. Joe Spano as Sgt. Henry Goldblume

Instead of acting, Blacque went on to adopt ten children and ultimately became the national adoption spokeswoman under President George H.W. Bush's presidency.

"Officer J.D. LaRue" is a skilled undercover officer most known for his drinking, womanizing, and get-rich-quick schemes. His Captain frequently confronts him about his persistent drinking, which nearly led to his dismissal from the force. Kiel Martin, the performer, was almost as unfortunate as his character. Martin had three marriages and three divorces. He, like his character on Hill Street Blues, struggled with drinking but overcame it. Martin died at the age of 46 from lung cancer at his home in Rancho Mirage, California.

"Sgt. Lucy Bates" was one of the few characters in Hill Street Blues that had a true narrative throughout the show's run. Despite being considered "one of the lads," she may be judgemental of her coworkers at times. She is generally paired with "Joe Coffey," with whom she has an ambivalent connection. Betty Thomas was offered the opportunity to direct select episodes of Hooperman after many lesser parts in other TV and film productions, which led to a mid-ranged directorial career. She directed a number of television movies, including My Breast, That Guy, and The Loop, as well as feature films such as Dr. Dolittle, I Spy, and John Tucker Must Die.

Officer Andrew Renko is the partner of "Officer Bobby Hill" and is originally from New Jersey. He claims to never venture west of Chicago and talks with a southern drawl while listening to country music. Despite the fact that he is known to make the occasional racist insult, he considers his companion "Bobby" to be one of his closest pals. Charles Haid went on to appear in other TV programs, including Third Watch. Haid has also appeared in animated movies such as The Get Along Gang and Home On The Range. He began directing, like some of his former co-stars and has worked on shows such as Buffalo Soldier, NYPD Blue, Nip/Tuck, Sons of Anarchy, and Breaking Bad.

Throughout the show, "Joyce Davenport" is a very successful public attorney who is in a relationship with "Capt. Furillo." Their connection is kept concealed until the show's second season. Veronica Hamel received five Emmy nominations for her work in Hill Street Blues. Following that, she appeared in Alan Alda's A New Life, as well as other films such as Taking Care of Business. "Officer Joe Coffey" is a typical patrol officer that works alongside "Lucy Bates" throughout the show. He is one of the larger cops on the team and is generally assigned to handle the heavy lifting. Ed Marinaro was a professional football player before pursuing acting roles in a variety of programs. He first appeared on television in series such as Laverne & Shirley and Sisters. His two most memorable parts were in the Spike TV comedy Blue Mountain State and Hill Street Blues. In May 2015, he was chosen as Tuff TV's first celebrity spokesperson.

"Fay Furillo," "Frank's" ex-wife, appeared multiple times at the start of the series. She was usually seen bugging "Frank" about alimony, child support, and her own personal difficulties. "Frank" eventually gets enough and tells her to leave him alone because they are no longer married. She later falls in love with a couple of the cops on "Frank's" task group. Barbara Bosson married Hill Street Blues creator Steven Bochco, who also developed several of the shows she eventually starred in, including L.A. Law, Murder One, and Cop Rock. Before divorcing in 1997, the couple had two children.
